Comparative Study between the Effect of Atorvastatin and Naltrexone on Hepatic Fibrosis Induced by Bile Duct Ligation in Rats

چکیده
Hepatic fibrosis is one of the common pathological features of chronic liver disease. Atorvastatin and naltrexone previously showed a beneficial effect on hepatic fibrosis. In the present study, we compared the effect of atorvastatin and naltrexone on a rat model of hepatic fibrosis induced by BDL. Methods: This study was carried out on 4 groups each of 10 albino rats; Group 1: Sham operated group, Group 2: BDL group + No treatment, Group 3: BDL + Atorvastatin orally for 4 weeks and Group 4: BDL + Naltrexone SC for 4 weeks. Serum bilirubin, ALT, AST, ALP and hyaluronic acid and hepatic hydroxyproline content, GSH, GSH/GSSG ratio and TNF-α level were measured. Histopathological examination of the liver tissue was performed. Results: Treatment with either atorvastatin or naltrexone showed significant increase in hepatic GSH and hepatic GSH/GSSG ratio with significant decrease in serum hyaluronic acid and hepatic hydroxyproline content and TNF-α and improvement of histopathological picture of hepatic tissue. Also, naltrexone produced significant decrease in serum total bilirubin, AST, ALT and ALP while atorvastatin showed significant increase in their levels. Moreover, treatment with naltrexone showed significant reduction in serum hyaluronic acid level and more improvement in the histopathological picture of hepatic tissue than atorvastatin. Conclusion: Naltrexone is more effective than atorvastatin in attenuation of BDL induced hepatic fibrosis and both could be of beneficial effects in treatment of liver fibrosis in clinical practice. The preventive effect of atorvastatin on liver fibrosis in the bile duct ligation rats via antioxidant activity and down-regulation of Rac1 and NOX1
Objective(s): Atorvastatin is a cholesterol-lowering agent capable of inhibiting 3-hydroxy-3-methylglutaryl coenzyme A reductase. Recent studies have demonstrated new facets of atorvastatin, such as antioxidant and anti-fibrotic properties. We investigated the effect of atorvastatin on hepatic injury via the measurement of the antioxidant capacity and protein expressio...
متن کاملEffect of captopril on TNF-α and IL-10 in the livers of Bile Duct ligated Rats
Background: The renin-angiotensin system has an important role in hepatic inflammation and fibrosis. Renin-angiotensin system blockade by angiotensinconverting enzyme (ACE) inhibitors provides some protective effects against hepatic fibrogenesis. Captopril as an ACE inhibitor can decrease inflammatory mediators and attenuate hepatic fibrosis in the livers of bile duct ligated (BDL) rats. Object...

Objective (s): The anti-fibrotic effect of chronic berberine (BBR) had demonstrated previously in a rat model of bile duct ligation (BDL). The aim of present study was to investigate hepatoprotective effect of BBR nanomicelles on liver cirrhosis induced by BDL in male rats.Materials and methods: After 21 days of drugs’ treatments, the serum and tissue levels of hepatic markers were measured and...

Aims: Cholestasis is a type of liver disease due to structural damage and dysfunction of hepatobiliary system which at first, results in accumulation of bile acids and other toxins in plasma and hepatic tissue. The aim of the current study was to investigate the possible hepatoprotective effects of rosmarinic acid against oxidative stress and liver injury in bile duct ligation (BDL)- induced ch...

Objective(s): We aim to examine possible ammonia lowering effects of mesoporous silica SBA-15 in rats after the common bile duct ligation (BDL). We also evaluate the effect of SBA-15 treatments during 28 days of BDL on locomotion and rearing behavior, as well as on the gene expression of Jnk3 and p38alpha (p38α) mitogen-activated protein kinases in the prefrontal corte...
